Tacrolimus 8-epimer (Tacrolimus EP impurity D) is the C-8 epimer and degradation impurity of Tacrolimus (HY-13756). Tacrolimus 8-epimer forms via base-catalyzed epimerization of Tacrolimus, and also generates under iodine-catalyzed free radical conditions. Tacrolimus 8-epimer is mainly used as a reference substance for impurity analysis, pharmaceutical quality control and stability-related studies of Tacrolimus.

Tacrolimus 8-epimer (Tacrolimus EP impurity D) (compound 2) can be prepared via DBN (1,5-diazabicyclo[4.3.0]non-5-ene; 302 mg; 2.4 mmol)-catalyzed epimerization of Tacrolimus (1 g; 1.2 mmol) in dichloromethane (CH2Cl2). The reaction system is slowly warmed from 0°C to room temperature and stirred overnight. After purification by preparative reverse-phase HPLC, 147 mg of white amorphous Tacrolimus 8-epimer is obtained with a yield of 14.7%, and crystals suitable for single-crystal X-ray diffraction analysis are obtained via crystallization[2].
Tacrolimus 8-epimer is a transformation product that may form during the production, formulation preparation or storage of Tacrolimus. The prepared Tacrolimus transformation product is mainly used as a reference substance in pharmaceutical impurity analysis, and assists in the identification of Tacrolimus degradation impurities via methods such as HPLC, NMR and mass spectrometry[3].

Select the appropriate dissolution method based on your experimental animal and administration route.
- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (3.11 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
