Trichlorfon ethyl
Trichlorfon ethyl is an analog of Trichlorfon (HY-B1220), as well as an inhibitor of Butyrylcholinesterase and Bacillus subtilis Esterase. Trichlorfon ethyl acts as a prenatal brain growth inhibitor, causing mild reductions in total brain weight, medulla oblongata weight and tectum weight in newborn guinea pigs, without affecting cerebellar weight. Trichlorfon ethyl does not induce severe prenatal brain hypoplasia in guinea pig offspring. Trichlorfon ethyl can be used in studies related to brain hypoplasia.

Trichlorfon ethyl (Compound Ih) (0.1 mM; 1-10 min) inhibits butyrylcholinesterase from horse blood serum with a kII of 2.75×103 L mol-1 min-1 and Bacillus subtilis esterase with a kII of 4.06×102 L mol-1 min-1[2].

Result:Showed no significant reductions in total brain weight or weights of cerebellum, medulla oblongata, diencephalon, hippocampus, quadrigemina, or cerebral cortex in offspring exposed to 125 mg/kg, compared to saline controls.
Showed a significant reduction in total brain weight (mean 2.47 g, p < 0.05), medulla oblongata weight (mean 163 mg, p < 0.05), and quadrigemina weight (mean 73 mg, p < 0.05), but no significant reduction in cerebellum weight in offspring exposed to 138 mg/kg (day 42) and 121 mg/kg (day 44).
Reduced red blood cell acetylcholinesterase activity to 51% of control levels 24 hours after administration of 125 mg/kg.
Observed no significant changes in glutamate decarboxylase, choline acetyltransferase, or acetylcholinesterase activities in cerebellum, medulla, or cerebral cortex of offspring exposed to ethyl-trichlorfon.
