Description

Ac-WLA-AMC is a fluorogenic substrate of caspase-3. Ac-WLA-AMC is cleaved to release the fluorescent moiety 7-amino-4-methylcoumarin (AMC), which can be used to quantify the β5c subunit activity[1][2].

In Vitro

Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs). Labeling of caspase: 1.Cells were harvested and washed with PBS. 2.After centrifugation, they were resuspended in PBS at a concentration of 2 × 106 cells/100 μl. 3.25 μl of the suspension was added to a 96-well plate and mixed with the appropriate peptide substrate dissolved in a standard reaction buffer (100 mM HEPES, 10 % sucrose, 5 mM DTT, 0.001 % NP-40, and 0.1 % CHAPS, pH 7.2). 4.Ac-DNLD-AMC was monitored by AMC liberation in a VarioScan Flash multimode detector (Thermo Scientific) using 380-nm excitation and 460-nm emission wavelengths.
