Methyl yellow (Solvent Yellow 2) is a pH indicator. Methyl yellow appears red at low pH, and becomes yellow above pH 4.0^[1].

Induced Disease Models

Methyl yellow can be used in animal modeling to create hepatic tumor models. After administration of Methyl yellow, the levels of dihydroxycholan acid (DHA) decrease in rats that secrete more than 10 mg/100 g body weight of trihydroxycholan acid (THA) daily. The bile volume also increases in the treated rats, and there is a strong correlation between DHA levels and bile volume^[2].

Induction of liver tumor^[2]

Background

Methyl yellow can degrade cholesterol into bile acids, and specific disorders in bile acid metabolism may lead to the generation of endogenous carcinogens, which can induce liver tumors.

Specific Modeling Methods

Rat: G.G. strain • male • 3-4 months old
Administration: 0.06 % of diet • feed • 2-5 months

Note

(1) Create a fistula in the rat's bile duct, and collect bile from days 2 to 4 after cannulation, discarding the bile from the day before death.
(2) The rats were either killed after a period of bile discharge or euthanized with ether. A histological examination was performed on the livers of the euthanized mice.

Modeling Indicators

Phenotypic observation: Increased bile production and bile drainage lead to bile duct hyperplasia and liver necrosis; the survival time of rats after bile duct cannulation is shortened, and they experience significant weight loss.
