Progoitrin potassium

Progoitrin potassium is an orally active glucosinolate. As the precursor of goitrin (HY-N0224A), an antithyroid compound, progoitrin releases goitrin via hydrolysis by myrosinase or gastrointestinal glucosidase. Progoitrin converts to goitrin in vivo and exerts antithyroid activity by inhibiting the accumulation of radioactive iodine in the thyroid gland. Progoitrin potassium can be used in the research of Graves' disease (thyrotoxicosis).

Potassium progoitrin (1.4 μmol/g; 5-60 min at 150-200 °C) shows the lowest thermal stability in pH 9.0 phosphate buffer and undergoes thermal degradation to produce volatile flavor compounds^[1].

In Vivo

Progoitrin (potassium) (20-1000 mg; p.o.; s.c.; i.p.; 1 to 23.5 hours before I¹³¹ injection) exhibits dose-dependent antithyroid activity in male Holtzman rats, with oral administration generally more potent than subcutaneous or intraperitoneal routes at equivalent doses, reducing thyroidal I¹³¹ uptake to as low as 31.8% of control values^[3].
