BChE/MAO-B-IN-6 is a dual inhibitor with inhibitory activity against both butyrylcholinesterase (BChE) and monoamine oxidase B (MAO B), with an IC50 of 557 nM against BChE and an IC50 of 142 nM against MAO B. BChE/MAO-B-IN-6 reduces IL-6 release from stimulated microglia and antagonizes 6-hydroxydopamine (HY-B1081) toxicity in neuroblastoma cells. BChE/MAO-B-IN-6 can be used in the research of neurodegenerative diseases[1].

In Vitro

BChE/MAO-B-IN-6 (Compound 9) (0.1-5 μM; 1-3 h) attenuates 6-OHDA-induced cytotoxicity in human neuroblastoma SH-SY5Y cells in a dose-dependent manner[1].
BChE/MAO-B-IN-6 (0.01-1 μM; 24 h) reduces IL-6 release in LPS/IFN-γ/H2O2-activated human microglial HMC3 cells in a dose-dependent manner, with significant inhibitory effects at concentrations of 0.1 μM and 1 μM[1].

Cell Viability Assay[1]

Cell Line: SH-SY5Y human neuroblastoma cells
Concentration: 0.1, 0.5 and 5 μM
Incubation Time: 1 h or 3 h (pre-incubation)
Result: Significantly attenuated 6-OHDA-induced cytotoxicity at all tested concentrations; the highest protection was observed at 0.5 μM, with viability levels comparable to a combination of tacrine and safinamide at equivalent total concentration (0.1 μM total).
