BL20-MMAE is an antibody-drug conjugate targeting ROR1, which is formed by conjugating an anti-ROR1 antibody with the Auristatin payload MMAE (HY-15162) via a BL20 linker[1].

In Vitro

BL20-MMAE (10 mg/mL; maintained at 28°C, analyzed after conjugation) conjugates with ROR1-4 antibody to form a purified ADC with a precise drug-to-antibody ratio of 4.0[1].
BL20-MMAE (10 mg/mL; maintained at 28°C, analyzed after conjugation) conjugates with ROR1-12 antibody to form a purified ADC with a precise drug-to-antibody ratio of 4.0[1].

In Vivo

BL20-MMAE (administered as ROR1-12-BL20E) (5 mg/kg; i.v.; on day 0 and day 4) achieves 92% tumor growth inhibition in JeKo-1 leukemia xenografts, with partial regression observed in 1 of 6 mice[1].
BL20-MMAE (administered as ROR1-12-BL20E) (5 mg/kg; i.v.; once weekly; 3 doses) achieves 49% tumor growth inhibition in MDA-MB-231 breast cancer xenografts[1].
BL20-MMAE (administered as ROR1-4-BL20E) (5 mg/kg; i.v.; once weekly; 3 doses) achieves 39% tumor growth inhibition in MDA-MB-231 breast cancer xenografts[1].
BL20-MMAE (administered as ROR1-12-BL20E) (5 mg/kg; i.v.) achieves 51% tumor growth inhibition in H1975 non-small cell lung cancer xenografts[1].
BL20-MMAE (administered as ROR1-4-BL20E) (5 mg/kg; i.v.) achieves 69% tumor growth inhibition in H1975 non-small cell lung cancer xenografts[1].

Animal Model: tumor-bearing mice (implanted with JeKo-1 cells, tumors grown to ~100 mm3)[1]
