Aur

Three Aurora kinases police chromosome segregation. Aurora A is centrosomal and is activated by binding TPX2; it drives centrosome maturation, spindle assembly and mitotic entry, phosphorylating substrates such as TACC3 and NEDD1. Aurora B is the catalytic subunit of the chromosome passenger complex with INCENP, survivin and borealin, and phosphorylates histone H3 Ser10, MCAK and kinetochore components to destabilize erroneous microtubule attachments while supporting checkpoint signaling and cytokinesis. Aurora C is a meiosis-enriched paralog that partly substitutes for Aurora B. All three are overexpressed in tumors, and inhibitors such as alisertib, barasertib and hesperadin have been evaluated in oncology trials.
