Anticancer agent 312
Anticancer agent 312 is a heterogeneous nuclear ribonucleoprotein A18 (hnRNP A18) inhibitor with an IC50 of 2.9 μM. Anticancer agent 312 binds to the RNA recognition motif of hnRNP A18 and blocks its binding to homologous RNA transcripts. Anticancer agent 312 acts as a protein synthesis inhibitor by blocking hnRNP A18-mediated mRNA translation. Anticancer agent 312 is applicable to research on cancers such as melanoma.

Description
In Vitro
Anticancer agent 312 (Compound 5) (3 h) binds to and stabilizes hnRNP A18 in LOX-IM-VI melanoma cells, with an EC50 of 23.8 μM[1].
Anticancer agent 312 disrupts hnRNP A18-mediated transcript stability and translation in LOX-IM-VI melanoma cells, and reduces the expression of CAT protein carrying the Trx 3'UTR reporter gene[1].
Anticancer agent 312 (0-200 μM) downregulates the protein expression of thioredoxin in LOX-IM-VI melanoma cells in a dose-dependent manner[1].
Anticancer agent 312 (0-200 μM) downregulates CTLA-4 protein expression in PC-3 prostate cancer cells in a dose-dependent manner[1].
Anticancer agent 312 (0-150 μM; 24 h) selectively reduces the viability of various human cancer cell lines, and exhibits moderate toxicity to normal human mammary epithelial cells at high concentrations[1].
