DC-peptoid-1
DC-peptoid-1 is a specific binder and crosslinker targeting the phosphorylated Brd4 PDID domain, with a dissociation constant (Kd) of approximately 50-100 μM for human-derived targets. DC-peptoid-1 only binds to phosphorylated PDID and fails to recognize the non-phosphorylated form or other domains (such as Brd4 598-785). DC-peptoid-1 effectively crosslinks with the target protein both in solution and cell lysates. It can successfully capture phosphorylated PDID from complex systems via immobilization, without binding to bacterial-derived non-phosphorylated proteins or other non-specific phosphoproteins. DC-peptoid-1 has the potential to serve as a phosphoprotein-specific antibody substitute for applications such as immunoaffinity purification.

Description
In Vitro
Soluble DC-peptoid-1 binds to the phosphorylated PDID of Brd4 protein, with an estimated K0 of 50-100 μM, and shows extremely weak binding ability to unmodified PDID[1].
Cys-DC-peptoid-1 selectively captures the phosphorylated PDID domain of Brd4 protein from Sf9 cell lysates, but does not bind to unmodified PDID, and exhibits certain non-specific binding to other components in the lysates[1].
BD-DC-peptoid-1 (biotin-labeled, 0.5 μM) specifically cross-links with 0.5 μM phosphorylated Brd4 protein in solution in the presence of 2 mM NaIO4[1].
BD-DC-peptoid-1 (0.5 μM) cross-links with phosphorylated Brd4 protein in intact Sf9 cell lysate in the presence of 2 mM NaIO4, but it also undergoes non-specific cross-linking with other proteins in the lysate[1].
