MBD (7-(p-Methoxybenzylamino)-4-nitrobenz-2,1,3-oxadiazole) is a hydrophobic fluorescent probe that is used to detect conformational changes of the complement subcomponent C1r during its autoactivation process. MBD acts by selectively binding to exposed hydrophobic regions on the C1r macromolecule. The detection wavelength of MBD in the free unbound state is Ex/Em = 490/560 nm, whereas the maximum emission wavelength of MBD upon binding to C1r exhibits a blue shift of approximately 20 nm relative to 560 nm.

Description
In Vitro
1. Stock Solution Preparation[2]
1.1 Solvent: DMSO.
2. Working Solution Preparation
2.1 Diluent: 20 mM Tris-HCl buffer containing 0.15 M NaCl.
2.2 Working concentration: The MBD concentration in the final assay system is 4 μM.
2.3 Note: Adjust the working solution concentration as needed; prepare fresh before use.
3. Staining Procedure
3.1 Sample type: Purified C1r protein sample at a concentration of approximately 40 μg protein/mL.
3.2 Incubation conditions: Perform the binding assay using 4 μM MBD working solution at 20°C and pH 7.4.
5. Detection and Analysis
5.1 Instrument: Spectrofluorometer.
5.1.1 Excitation/emission wavelengths: Set the excitation wavelength to 490 nm and record the fluorescence emission spectrum in the range of 500-600 nm.
5.2 Result analysis:
5.2.1 Free MBD exhibits maximum fluorescence emission at 560 nm; upon addition of proenzyme C1r, the fluorescence intensity increases significantly, and the maximum emission wavelength shifts by approximately 20 nm toward shorter wavelengths.
5.2.2 Upon addition of activated C1r, the fluorescence spectrum of MBD shows no significant change, indicating that proteolytic activation disrupts the hydrophobic region in proenzyme C1r that binds MBD.
