CD367/CLEC4A

CD367, or DCIR (CLEC4A), is an inhibitory C-type lectin receptor of the DC-SIGN subfamily expressed on dendritic cells and macrophages. Its hallmark is a cytoplasmic ITIM that recruits phosphatases to restrain signaling from activating receptors, positioning DCIR as a negative regulator of antigen-presenting-cell function. The extracellular lectin domain binds high-mannose glycans and has been shown to capture HIV-1 gp120, promoting viral attachment and trans-infection of T cells; it also interacts with vimentin exposed on certain cells. Because of these properties, recombinant CD367/CLEC4A is valuable for HIV-entry studies, glycan-binding analysis, and research on inhibitory lectin checkpoints in innate immunity.
