YO-PRO-1 is a non-cell-membrane-penetrating green fluorescent dye. YO-PRO-1 cannot penetrate normal cell membranes but can penetrate the membranes of apoptotic and necrotic cells. YO-PRO-1 has a high affinity for DNA. When it is not bound to DNA, it emits almost no fluorescence, and when it binds to DNA, it can emit green fluorescence (Ex/Em: 488/530±30 nM). YO-PRO-1 is often used together with Propidium Iodide (HY-D0815) to analyze and identify apoptotic and necrotic cells.

Simple steps for YO-PRO-1 staining: Prepare cell/tissue samples; Wash the samples after incubating with YO-PRO-1 for 30 minutes to 1 hour. It can be co-stained with Hoechst 33342 (HY-15559), Propidium Iodide (HY-D0815), etc.; Microscopic observation and detection: Image by excitation with a 488 nm laser line[1][2].

Annexin V plus membrane-impermeant dye apoptosis staining
Annexin V-based apoptosis assays rely on the detection of phosphatidylserine (PS) externalization from the inner leaflet of the plasma membrane to the outer leaflet, an early biochemical hallmark of apoptosis. Fluorescently labeled Annexin V binds PS in a calcium-dependent manner, enabling identification of early apoptotic cells by flow cytometry or fluorescence microscopy. When combined with a membrane-impermeant DNA-binding dye (e. g. , propidium iodide), this approach allows discrimination between viable (Annexin V−/dye−), early apoptotic (Annexin V+/dye−), and late apoptotic or necrotic (Annexin V+/dye+) cell populations by assessing membrane integrity and PS exposure.

Apoptosis is a regulated, generally non-lytic cell-death pathway that removes unwanted, damaged, infected, or abnormal cells through coordinated morphological changes, caspase activation, DNA fragmentation, and membrane remodeling.
