Velaresol (BW 12C) is a small molecule oxyhemoglobin stabilizer. Velaresol stabilizes hemoglobin’s oxy-conformation, shifts oxygen dissociation curves left, induces tumor hypoxia, alters radiosensitivity, protects normal tissues from radiation damage, and reduces sickled cell counts. Velaresol can be used for the research of cancer and sickle cell disease.

Biological Activity
Velaresol (5 mM; 30 min pre-incubation under air) increases the O2 affinity of Hb in human HbA red blood cells, reducing the P02 required for half-maximal O2 saturation to 7 mmHg[4].
Velaresol (5 mM; 30 min pre-incubation under air) preserves KCC1 activity at low oxygen tensions in human HbA red blood cells, uncoupling transporter function from both oxygen tension and haemoglobin O2 saturation[4].
Velaresol (1.5-5.0 mM; 30 min) increases erythrocyte deformability and reduces sickled cell formation in washed erythrocytes from homozygous SS, S/C, S/β0-thalassaemia, and S/hereditary[5].

Velaresol (70 mg/kg; i.p.; single dose) induces transient reductions in tumor blood flow and hemoglobin oxygen affinity in mice with rhabdomyosarcoma[2].
Velaresol (70 mg/kg; i.p.; single dose) alters hemoglobin oxygen affinity in WHT mice but has no significant effect on Fib/T tumor blood flow or radiosensitivity[2].
Velaresol (70 mg/kg; infusion) reduces relative tumour tissue perfusion, muscle tissue perfusion, while increasing skin blood flow in mice bearing RIF-1 tumors[6].

- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

This protocol yields a clear solution of ≥ 2.08 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (20.8 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.08 mg/mL (8.73 mM); Clear solution
This protocol yields a clear solution of ≥ 2.08 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (20.8 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
