TTX-030 (Eltivutabart) is a monoclonal antibody that allosterically inhibits CD39 ectonucleotidase activity, preventing the conversion of ATP to AMP. TTX-030 acts as a non-competitive allosteric CD39 ATPase inhibitor, binding to an N-terminal epitope distant from the active site and blocking nucleotide hydrolysis. TTX-030 has an IC50 of 0.20 nM, a Kd of 1.5 nM, and a Ki of 0.5 nM for human CD39. TTX-030 enhances CD4+ and CD8+ T cell proliferation, Th1 cytokine production, T cell function, dendritic cell maturation, and inflammasome activation. TTX-030 can be used in research on human B cell lymphoma and cancer.

In Vitro
TTX-030 (60 min) non-competitively inhibits CD39 ATPase in SK-MEL-28 cells with a Ki of 0.50 nM and a maximum inhibition rate of up to 85%, and maintains its inhibitory effect under washing, repeated ATP addition, and pH 6.5 conditions[2].
TTX-030 inhibits CD39-mediated Pi release in primary human B cells[2].
TTX-030 (200 ng/mL; 30 min) inhibits SK-MEL-28 CD39-mediated sequential hydrolysis of 3HATP to adenosine[2].
TTX-030 (0.5 μg/mL; 45 min) almost completely blocks CD39-mediated ADPase activity in human tonsils[2].
TTX-030 (15 µg/mL; 30 min) binds cell surface CD39 with 100-500 pM affinity, selectively recognizes human and cynomolgus monkey CD39, and localizes to an allosteric epitope at E142-Y159[2].

In Vivo
TTX-030 (250 μg per dose; intraperitoneal injection; days 5, 8, 11, and 14; four doses in total) combined with Pembrolizumab (HY-P9902) and autologous EBV-specific T cells exhibits stronger inhibition of LCL043 tumor growth than either antibody combined with T cells[1].

Result:Was without effect against LCL039 tumors as monotherapy.
Autologous EBV-specific T cells were more active alone in this setting; the antitumor activity of autologous EBV-specific T cells was enhanced.
At endpoint day 23, increased the percentage of tumor-infiltrating CD3+ cells and the number of tumor-infiltrating CD3+CD8+ T cells compared with cIg control mice.
Decreased CD39 expression levels on intratumor CD8+ T cells compared with cIg control.
PD-1 expression was increased on CD8+ T cells after injection; PD-1 expression levels on intratumor CD8+ T cells were reduced compared with cIg control.
IHC on day 23 LCL039 tumors showed increased CD8+ T-cell infiltration and reduced CD39 expression in the EBV-specific T-cell combination tumors.
