CN045
CN045 is a blood-brain barrier-permeable, selective M1 muscarinic receptor ligand with an IC50 value of 1 μM. CN045 enhances remyelination in mouse brain regions. CN045 can be used in studies related to multiple sclerosis.

Biological Activity
|
mAChR1 1 μM (IC50) |
CN045 (5 days) potently promotes differentiation of primary mouse PLP-EGFP transgenic oligodendrocyte progenitor cells in vitro with an EC50 of 40 nM, yielding a higher percentage of mature oligodendrocytes than Clemastine (HY-B0298)[1].
CN045 (5 days) potently stimulates differentiation of primary mouse OPCs into mature, myelin protein-expressing oligodendrocytes with an EC50 of 0.04 μM[2].
CN045 (1 μM; 14 days) promotes myelin-like ensheathment of engineered nanofibers by primary mouse OPCs, increasing both oligodendrocyte numbers and ensheathment length[2].

Result:Increased remyelination in the corpus callosum, hippocampus, and cerebral cortex compared to vehicle treatment.
Increased the number of PPD+ myelinated axons per unit area in the corpus callosum.
Increased the percentage area of PLP staining in the hippocampus.
Increased the percentage area of PLP staining in the cerebral cortex.
