Denecimig (Mim8) is a factor VIII-mimetic bispecific antibody with micromolar human binding affinity for Factor X and Factor IXa. Denecimig binds Factor X and Factor IXa, localizes both to the phospholipid surface, enhances the Factor IXa-mediated activation of Factor X to Factor Xa, and stimulates the proteolytic activity of Factor IXa via its monovalent anti-Factor IXa arm. Denecimig is applicable to research related to hemophilia A.

Denecimig efficiently enhances FX activation by localizing FIXa and FX to phospholipid surfaces, with its anti-FIXa arm alone increasing FIXa proteolytic activity 15,000-fold, and binds FIXa and FX with micromolar dissociation constants[1].
Denecimig (0.1-1000 nM) normalizes thrombin generation in congenital HA plasma with 15-fold greater potency than emicizumab[1].
Mim8 exhibits procoagulant activity in hemophilia A-like blood that is ~13-fold more potent than a sequence identical analog of emicizumab, as measured by peak thrombin generation[2].
