DOTAP methylsulfat is a cationic lipid reagent, a cationic derivative of trimethylammonium, linked to two 18-carbon fatty acid tails, each with a single olefin group. DOTAP methylsulfat can self-assemble with negatively charged ions (such as DNA) to form complexes, which can be adsorbed to the cell membrane surface and enter the cell by electrostatic interaction and endocytosis, respectively. DOTAP methylsulfat promotes endosomal membrane fusion with its own hydrophobic domain, releases DNA into the cytoplasm, and exerts gene delivery function. DOTAP methylsulfat can be widely used in research fields such as gene therapy, cell transfection, and non-viral vector design.

    Description

    DOTAP methylsulfat is a cationic lipid reagent, a cationic derivative of trimethylammonium, linked to two 18-carbon fatty acid tails, each with a single olefin group.     In Vitro

    Different mass ratios of DOTAP methylsulfat (different mass ratios; 4 hours transfection + 24 hours culture) and DOPE (HY-112005) have different optimal transfection efficiencies. In Huh7 and AGS cells, the optimal ratio is T1P0 (1:0)/T3P1 (3:1), and in A549 cells, the optimal ratio is T1P1 (1:1)/T3P1 (1:3), which is independent of liposome size or zeta potential[1].
    DOTAP methylsulfat mixed with lipids in certain ratios (e.g., 1:2, 1:1, 2:1) is more efficient than DOTAP methylsulfat alone in transfecting DNA, and still maintains transfection activity at 80% serum concentration. And the optimal transfection ratio of DOTAP methylsulfat to DNA is 12:1[2].

    DOTAP methylsulfat (30 min DNase treatment) DNA-lipid complexes remain stable to DNase I digestion in the presence of serum, and the addition of cholesterol does not affect the structural integrity of the complex[2].

    DOTAP methylsulfat (containing 1% NBD-DOTAP methylsulfat label; 2 h transfection) in flow cytometric analysis, cholesterol-modified liposomes bind to cells at a significantly higher rate in serum than pure DOTAP methylsulfat liposomes[2].

    Select the appropriate dissolution method based on your experimental animal and administration route.

    For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
    To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for in vivo experiments, it is recommended to prepare freshly and use it on the same day.
    The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

    • Protocol 1

      Add each solvent one by one:  10% EtOH    90% Corn Oil

      Solubility: ≥ 2.5 mg/mL (3.23 mM); Clear solution

      This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown). If the continuous dosing period exceeds half a month, please choose this protocol carefully.
