Etalanetug  (Synonyms: E2814)

Etalanetug (E2814) is a humanized high-affinity IgG1 antibody that targets tau protein and can cross the blood-brain barrier. Etalanetug inhibits the spread of pathological tau protein through high-affinity binding to the microtubule-binding region (MTBR). Etalanetug can be used in research related to Alzheimer's disease^[1][2].

Etalanetug (2 μg/mL) binds to recombinant human wild-type 2N4R tau protein with a high affinity of 88 pK_d^[2].
Etalanetug binds to recombinant human 2N3R tau protein in a dose-dependent manner, with slightly reduced affinity relative to 2N4R tau^[2].
Etalanetug (8.3 μM; 30 min pre-incubation, 6 days aggregation) potently inhibits heparin-induced aggregation of both recombinant wild-type and P301S mutant 2N4R tau protein in vitro^[2].
Etalanetug (0.3-10 μg; 48 h at 37 °C under 5% CO₂) immunodepletes tau seeds (K18 fibrils and full-length P301S tau monomer) in a dose-dependent manner, reducing intracellular tau deposition in Lenti-X 293 T cells overexpressing P301S 0N4R tau^[2].

  Immobilized Fetal-tau/0N3R Protein, Human (His, solution, HY-P73618) can bind Etalanetug. The EC₅₀ for this effect is 1.246 ng/mL.

  Flow cytometric analysis of 1X10⁶ SH-SY5Y cells labeling Tau with Etalanetug (HY-P990995, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/200 for an hour at 4℃. AF488-conjugated Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa Isotype Control (HY-P99001, blue) was used as the isotype control.
