GPC3 targeting peptide 2
GPC3 targeting peptide 2 is a glypican-3 (GPC3) targeting peptide that binds specifically to GPC3 with a Kd of 39.5 nM. GPC3 targeting peptide 2 can be conjugated to IRDye800 via a GGGSC linker for near-infrared fluorescence and photoacoustic imaging, with a tumor-to-adjacent liver fluorescence intensity ratio of approximately 2-fold. GPC3 targeting peptide 2 shows high specificity for immunofluorescence staining of HCC specimens. GPC3 targeting peptide 2 can be used in studies related to GPC3-targeted molecular imaging and early detection of hepatocellular carcinoma.

GPC3 targeting peptide 2 (ALLANHEELFQT) at 5 μM specifically binds to GPC3 on human Hep3B hepatocellular carcinoma cells; its binding level decreases significantly, by up to 8.7-fold, after GPC3 knockdown[1].
GPC3 targeting peptide 2 (0-200 μM; 30 min) competes with IRDye800-labeled GPC3 targeting peptide 2 for binding to human Hep3B hepatocellular carcinoma cells in a dose-dependent manner, confirming that this peptide mediates specific binding to GPC3[1].
GPC3 targeting peptide 2 (0-200 nM; 1 h) binds to human Hep3B hepatocellular carcinoma cells with high affinity, with a dissociation constant (Kd = 39.5 nM) and rapid binding kinetics (0.21 min−1)[1].
GPC3 targeting peptide 2 (5 μM; 10 min) specifically binds to human hepatocellular carcinoma (HCC) specimens in vitro, exhibits high sensitivity and specificity for distinguishing HCC from non-HCC and liver cirrhosis, and shows strong colocalization with anti-GPC3 antibodies[1].

Animal Model:NOD.Cg-Prkdcscid Il2rgtm1Wjl/SzJ (NSG) mice were obtained from Jackson Laboratory and bred by ULAM. Patient HCC specimens were cut into small pieces and implanted subcutaneously into NSG mice. Tumors were monitored biweekly for 4-6 weeks until ~1 cm, then harvested, sliced (~2×1×1 mm3), and implanted orthotopically in the liver.[2]
