IKKε-IN-1 is a IKKε inhibitor. IKKε-IN-1 reduces cell viability, inhibits colony formation and cell migration. IKKε-IN-1 induces autophagy (Autophagy) in cancer cells. IKKε-IN-1 can be used in the research of cancers including colorectal cancer, hepatocellular carcinoma, bladder cancer, breast cancer, lung cancer, and cervical cancer[1].

In Vitro

IKKε-IN-1 (6.25-50 μM; 72 h) reduces the viability of HCT116, HepG2, T24, MDA-MB-231, A549 and Hela cells, with the strongest activity against HCT116 colorectal cancer cells (IC50 = 13.37 μM)[1].
IKKε-IN-1 (6.25-50 μM; 2-14 days) inhibits the long-term proliferation of HCT116 and HepG2 cells, and reduces the migration of HCT116 cells[1].
IKKε-IN-1 (12.5-50 μM; 48 h) induces autophagosome formation but blocks autophagic flux in HCT116 cells[1].

Cell Cytotoxicity Assay[1]

Cell Line: HCT116、HepG2、T24、MDA-MB-231、A549、Hela cells
Concentration: 6.25、12.5、25、50 μM
Incubation Time: 72 h
Result: Reduced cell viability in all six cancer cell lines in a concentration-dependent manner.
Exhibited IC50 values of 13.37 μM (HCT116), 17.44 μM (HepG2), 29.733 μM (T24), 32.48 μM (MDA-MB-231), 66.3 μM (A549), and 68.05 μM (HeLa).

Cell Proliferation Assay[1]

Cell Line: HCT116, HepG2 cells

Incubation Time: 14 days
Result: Reduced colony formation in both cell lines in a concentration-dependent manner.

Cell Migration Assay [1]

Cell Line: HCT116 cells

Result: Inhibited the migration, had a more pronounced effect on reducing
cell migration at 12.5 μM.

Cell Autophagy Assay[1]

Cell Line: HCT116 cells
Concentration: 12.5、25、50 μM
Incubation Time: 48 h
Result: Increased the LC3B-II/LC3B-I ratio in a concentration-dependent manner, indicating increased autophagosome formation, and also increased p62 levels in a concentration-dependent manner, indicating blocked autophagic flux.
Significantly increased the number of yellow autophagosome puncta, confirming impaired fusion of autophagosomes and lysosomes.
