Isatin-O (Isatin β-oxime; Isatin 3-oxime) is an acetylcholinesterase (AChE) reactivator capable of crossing the blood-brain barrier. Isatin-O reactivates paraoxon-inhibited acetylcholinesterase in the central and peripheral nervous systems. As a glycosylation substrate, Isatin-O undergoes complete glycosylation in a Rauvolfia serpentina cell suspension culture system to form isatin-3-oxime-glucoside. Isatin-O can be used in studies related to organophosphate poisoning[1][2].

In Vitro

Isatin-O (10-100 μmol/L; 10-120 min) reactivates paraoxon-inhibited Electrophorus electricus acetylcholinesterase, reaching ~8% reactivation at 10 μmol/L within 10 min and ~23% reactivation at 100 μmol/L after 120 min, with better initial reactivation performance than 2-PAM at 10 μmol/L[1].
Isatin-O (5.76 mM; 72 h) is completely glucosylated by Rauvolfia serpentina plant cell suspension cultures to form Isatin-3-oxime-β-D-glucopyranoside within 72 h, with the glucoside accumulating intracellularly without further metabolism[2].
