JNJ-2594

JNJ-2594 is an inhibitor of dengue virus (DENV) NS5 RNA-dependent RNA polymerase (RdRP). JNJ-2594 inhibits the elongation activity of DENV2 and DENV3 NS5 polymerases with IC₅₀ values of 5.8 μM and 3.2 μM, respectively, and inhibits de novo RNA synthesis of DENV2 with an IC₅₀ of 23.5 μM. JNJ-2594 exhibits cross-inhibitory effects on human mitochondrial RNA polymerase. JNJ-2594 can be used in research on the discovery of dengue virus NS5 polymerase and DENV RdRP inhibitors.

JNJ-2594 (3 h) potently inhibits full-length DENV2 NGC NS5 polymerase activity with an IC₅₀ of 5.8 μM and full-length DENV3 SL NS5 polymerase activity with an IC₅₀ of 3.2 μM in a 3-hour biochemical elongation assay^[1].
JNJ-2594 binds to biotinylated DENV2 PG NS5 RdRp with a K_d of 10 μM and an R_max of 249 RU in an SPR assay at 20 °C^[1].
JNJ-2594 (1.2-1000 μM; 20 min preincubation, 20 min RNA synthesis incubation) inhibits de novo RNA synthesis by full-length DENV2 TVS01 NS5 with an IC₅₀ of 23.5 μM in a gel-based RNA synthesis assay^[1].
JNJ-2594 inhibits primed RNA synthesis by human mitochondrial RNA polymerase with an IC₅₀ of 5.2 μM in a gel-based RNA synthesis assay^[1].

Viral Infection

JNJ-2594 (100 μM; 3 days) shows no measurable antiviral activity against DENV2/eGFP (EC₅₀ > 50 μM in Vero cells, EC₅₀ > 61.7 μM in Huh7 cells) and no measurable cytotoxicity (CC₅₀ > 100 μM in Vero cells, CC₅₀ > 50 μM in Huh7 cells) in 3-day cell-based assays^[1].
