BPN-01
BPN-01 is a fluorescent probe targeting translocator protein (TSPO) and human epidermal growth factor receptor 2 (HER2) with binding affinities of -10.7 kcal/mol for TSPO and -11.3 kcal/mol for HER2. BPN-01 is promising for research of solid tumor imaging during fluorescence image-guided surgery (FIGS) (Ex/Em = 475/510 nm).

In Vitro
BPN-01 (3.91 μg/mL, 7.81 μg/mL, 15.63 μg/mL, 31.25 μg/mL, 62.5 μg/mL, 125 μg/mL, 250 μg/mL, 500 μg/mL; 48 h; B16-F10) maintains B16-F10 cell viability at ~100% across all concentrations[1].
BPN-01 (0.01 mM, 0.05 mM, 0.1 mM, 0.5 mM, 1.0 mM; 24 h; DU-145, B16-F10, C2C12) stains DU-145 and B16-F10 cancer cells at 0.01 mM but not at higher concentrations or in C2C12 cells[1].

Bioluminescent and fluorescent imaging xenograft models use tumor cells engineered to express optical reporters so tumor engraftment, growth, dissemination, and treatment response can be monitored longitudinally in living animals and validated ex vivo. Bioluminescence imaging usually measures luciferase activity after substrate administration and is commonly used as a surrogate for viable reporter-expressing tumor burden, while fluorescence imaging measures reporter or probe emission and can support tumor localization, ex vivo confirmation, or complementary multimodal analysis.
