GL67
GL67 (N4-Spermine cholesteryl carbamate) is a cationic lipid that serves as a non-viral siRNA delivery vector and a non-viral gene transfer vector. GL67 enhances the encapsulation efficiency of siRNA in liposomal formulations and promotes the cellular uptake of siRNA across cancer cell membranes. GL67 mediates the transduction of plasmid DNA into cells. GL67 can be used in drug delivery-related research.

Biological Activity
GL67 lipid complexes display enlarged particle sizes and positively shifted zeta potentials as the cationic lipid-to-siRNA molar ratio rises from 1:1 to 3:1, with the 100% GL67 formulation at 3:1 generating a high positive zeta potential of 47 mV. It elevates siRNA encapsulation efficiency across all tested ratios, and complete encapsulation is attained with less GL67 at higher lipid-siRNA ratios[1].
GL67 pentahydrochloride-based lipid complex at a 3:1 cationic lipid/siRNA molar ratio shows favorable metabolic and membrane integrity compatibility with A549 cells and boosts cellular uptake of Cy3-siRNA in A549 cells[1].
GL67 (GL-67) mediated markedly lower gene transfection efficiency in polarized differentiated primary human and canine tracheobronchial epithelial cells cultured at the air-liquid interface, along with suppressed cell division and elevated transepithelial electrical resistance. Its transfection capacity dropped with higher cell confluence in plate-grown unpolarized human bronchial epithelial cells and declined in growth-arrested, poorly proliferative primary human bronchial epithelial cells[2].

GL67:pDNA complexes (0.6 mM GL67 complexed with 3.6 mM pCF1-CAT; intranasal bolus (lungs); nasal perfusion) mediate >1000-fold higher gene expression than naked pDNA in the lungs of healthy BALB/c mice, but only minimal, statistically insignificant enhancement of gene expression in the nasal epithelium[2].
