Oliveroline

Oliveroline is an aporphine alkaloid with anticancer activity and a G₂ DNA damage checkpoint inhibitor. Oliveroline enables cells to escape G₂ phase arrest after DNA damage. Oliveroline (compound 1) (0.01-100 μM; 8 h post-irradiation) inhibits the G₂ DNA damage checkpoint in MCF-7 mp53 cells in a concentration-dependent manner^[1].
Oliveroline (24 h pre-irradiation) inhibits proliferation of MCF-7 mp53 cells with an IC₅₀ of 45 μM in the absence of DNA damage, and shows ~2.3-fold increased potency (IC₅₀ = 20 μM) in cells treated with 6.5 Gy ionizing radiation^[1].
Oliveroline inhibits the growth of human nasopharyngeal carcinoma (NPC-TW01), non-small cell lung carcinoma (NCI-H226), T cell leukemia (Jurkat), renal carcinoma (A498), lung carcinoma (A549), and fibrosarcoma (HT1080) cell lines, with the highest potency against A498 cells (IC₅₀ = 3.68 μM) and IC₅₀ values ranging from 3.68-13.28 μM across all lines^[2].
