Samrotamab (PR-1498487) is a humanized IgG1-κ chimeric monoclonal antibody targeting LRRC15, with a Kd of 5.42 nM for human LRRC15. Samrotamab can be used to synthesize antibody-drug conjugates (ADC). Samrotamab disrupts the LRRC15-SCG5 signaling module, binds to a membrane-proximal conformational epitope within the C-terminal leucine-rich repeat region of LRRC15, and binds to the lateral edge of the LRR solenoid while leaving the canonical concave β-sheet surface fully exposed. Samrotamab reduces the viability of bladder cancer cells, inhibits clonogenic growth, impairs cell migration, and compromises cell invasion. Samrotamab induces compensatory upregulation of LRRC15 transcripts while suppressing the expression of SCG5 mRNA. Samrotamab is applicable to research related to urothelial carcinoma, sarcoma, osteosarcoma, and undifferentiated pleomorphic sarcoma.

Samrotamab (PR-1498487) (20 µg/mL; 24 h) reduces viability, clonogenic growth, migration, and invasion of human T24 muscle-invasive bladder carcinoma cells, while altering transcript levels of LRRC15 and SCG5[1].
Samrotamab (8.7-250 nM; 120 s contact time, 600 s dissociation time) binds purified human LRRC15 ectodomain with high affinity (KD = 5.42 nM) and specificity[2].
Samrotamab (25 µM; 0, 6, 60, 600, 6000 sec labeling time, overnight incubation at 4 °C) recognizes a conformational epitope formed by clustered leucine-rich repeat elements within the C-terminal region (residues 361-369, 383-393, 402-417) of purified human LRRC15 ectodomain[2].
Samrotamab (0.98-31.25 nM; 120 s contact time, 600 s dissociation time) binding to purified human LRRC15 ectodomain requires the C-terminal region encompassing residues 360-402; deletion of residues distal to 426 does not impact binding[2].
Samrotamab (0.13 mg/mL) Fab binds the lateral edge of the C-terminal leucine-rich repeat region of purified human LRRC15 ectodomain at 2.63 Å resolution, leaving the canonical concave β-sheet surface exposed[2].
