Antcin A

Antcin A is an orally active modulator of p53 and glucocorticoid receptor (GR), with an IC50 of 8.55 μM against human GR. Through transcriptional activation of p53, Antcin A induces miR-200c and inhibits ZEB1, regulates epithelial-mesenchymal transition markers, and suppresses cancer cell migration/invasion. Antcin A activates GR, inhibits Na+/K+-ATPase and NLRP3 inflammasome assembly, pyroptosis (pyroptosis) and pro-inflammatory cytokine release, reduces hepatic lipid deposition, improves liver function, and reverses metabolic changes induced by the SARS-CoV-2 spike protein. Antcin A can be used in research related to breast cancer, head and neck cancer, non-alcoholic fatty liver disease, and coronavirus disease 2019 (COVID-19).

In Vitro
Antcin A (87.6-188.1 μM) selectively reduces cell survival in human TSCCa, GNM, and KB head and neck cancer cell lines with IC50 of 188.1 μM, 87.6 μM, and 179.6 μM, respectively[2].
Antcin A (0.1-10 μM; 1 h) induces glucocorticoid receptor nuclear translocation in human lung cancer A549 cells, with a minimal effective concentration of 10 μM for 1 h incubation[3].
Antcin A inhibits fMLP-induced superoxide generation in human peripheral neutrophils with an IC50 of 8.55 μM, exhibiting greater potency than ibuprofen[2].
Antcin A induces glucocorticoid receptor activation in human lung A549 cells, exerting an anti-inflammatory effect comparable to synthetic glucocorticoids via high-affinity binding to GR[2].
Antcin A (5-15 mg/L; 6 h pre-incubation, 12 h LPS/Nigericin treatment) dose-dependently inhibits pyroptosis and NLRP3 inflammasome activation in mouse liver Kupffer cells, reducing inflammatory factor secretion, membrane permeability, and cleavage of GSDMD-FL[4].
Antcin A (15 mg/L; 6-48 h) is non-toxic to mouse liver Kupffer cells, with no effect on cell viability or apoptosis over a 48-hour incubation period[4].
