IBI38D9-L is a human IgG1-type CD79b/CD3 bispecific T-cell engager. IBI38D9-L binds to human CD79b and CD3 with K_d values of 1.45 nM and 119 nM, respectively. IBI38D9-L mediates tumor cell recognition and T-cell engagement, activating T cells and inducing their proliferation and release of cytotoxic molecules. IBI38D9-L exerts T cell-mediated cytotoxicity in a tumor cell-dependent manner, thereby selectively depleting antigen-positive malignant B cells. IBI38D9-L can be used for research on B-cell non-Hodgkin lymphoma.

IBI38D9-L (0.5-80 nM; 180 s) binds human CD79a/b with a K_D of 1.45 nM and human CD3 with a K_D of 119 nM in SPR^[1].
IBI38D9-L (6 h) activates NFAT-luciferase reporter T cells in a CD79b-dependent manner, with EC₅₀ values of 0.034 nM in BJAB, 0.030 nM in Ramos, and 0.13 nM in WSU-DLCL2 cells^[1].

IBI38D9-L (1-3 mg/kg; days 8-22) significantly inhibited WSU-DLCL2 and Ramos tumor growth in human PBMC-reconstituted NOG mice^[1].
IBI38D9-L (3 mg/kg; single dose) increases tumor-infiltrating CD4⁺ and CD8⁺ T cells and activated T cells in Ramos tumors in humanized NOG mice^[1].

Adaptive Immunity

IBI38D9-L (3-10 mg/kg; days 0, 7, 14, and 21) depleted peripheral B cells and later T cells in human CD34⁺ HSC-engrafted NPG mice^[1].

Compared with control, IBI38D9-L increased tumor-infiltrating CD4⁺ and CD8⁺ T cells on both days 1 and 3 after administration.
Activated T cells (CD4⁺CD25⁺ or CD8⁺CD25⁺) were increased in tumor samples from the treated group.
Intracellular granzyme B and perforin within CD4⁺ and CD8⁺ T cells showed similar trends to CD25.
Immunofluorescence confirmed elevated CD3⁺ T cell infiltration in tumors from treated animals.

Animal Model:

HSC-NPG (female, engrafted with human CD34+ hematopoietic stem cells)^[1]
Dosage:

3 mg/kg and 10 mg/kg
Administration:

on days 0, 7, 14, and 21
Result:

Produced a large decrease in B cells in peripheral blood at day 3 after the first dose at 3 and 10 mg/kg, with almost no B cells detected on day 10 and thereafter.
No loss of CD3⁺ T cells was observed at day 3; however, almost no CD3⁺ T cells were detected at day 10 onwards.
