Naphthol AS-TR phosphate is a substrate for alkaline phosphatase and acid phosphatase. Naphthol AS-TR phosphate enables histochemical enzyme localization in bone sections. Naphthol AS-TR phosphate is hydrolyzed to release insoluble naphthol AS-TR, which couples with diazonium salts to form a locally deposited azo dye, used for histochemical staining of paraffin-embedded tissues and for localization in freeze-dried tissues.

Naphthol AS-TR phosphate disodium/Fast Red RC reagent (8-24 h) reveals that control 7TD1 B-cell hybridoma cells have high alkaline phosphatase (ALP) activity, Aphidicolin (HY-N6733)-treated 7TD1 B-cell hybridoma cells maintain intense ALP activity even during apoptosis, and Cycloheximide (HY-12320)-treated 7TD1 B-cell hybridoma cells show reduced ALP activity during apoptosis^[2].
