    Competitive ELISA results from mixing biotinylated HAstV1 spike at 125 nM with varying concentrations of Nipocalimab and adding these pre-mixtures to an FcRn-coated ELISA plate at pH 7.0. After washing, HRP-streptavidin was used to detect bound HAstV1 spike. Results are an average of three replicates, and error bars represent standard deviation. Each Nipocalimab concentration was compared to the 0 nM condition using a two-sided Welch's t test. Source data are provided as a source data file.

    • Immobilized Human FcRn/FCGRT&B2M Heterodimer protein, His can bind Nipocalimab. The EC50 for this effect is 8.97 ng/mL.
    • Flow cytometric analysis of 1.5×106 THP-1 cells labeling Nipocalimab (HY-P99037, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 lambda Isotype Control (HY-P99992, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
