Technical Support

Ozanezumab (GSK1223249) is a humanized, Fc-inactivated monoclonal antibody that targets the nervous system protein Nogo-A. Ozanezumab promotes neurite outgrowth and axonal regeneration by neutralizing Nogo-A signaling. Ozanezumab is used for research on neurodegenerative diseases such as amyotrophic lateral sclerosis and multiple sclerosis[1].

In Vitro

Ozanezumab reduces or reverses the inhibitory effect on axonal growth in a dose-dependent manner in cultured cerebellar granule neurons from postnatal rats[2].
Ozanezumab (0.7-100 μg/mL (4.67 to 667 nM); 1 h pre-incubation, 24 h total) dose-dependently neutralizes the inhibitory effect of GST-Nogo-A on axonal growth of cultured postnatal rat cerebellar granule neurons[3].
Ozanezumab exhibits specific immunohistochemical staining in conserved neural tissues, muscle tissues, and some other tissues in Rattus norvegicus (rat), Macaca (monkey), and Oryctolagus cuniculus (rabbit, central nervous system), while additional species-specific staining is present in neuronal tissues, pituitary tissues, and cardiac tissues of rodents and primates[3].

Biological Activity
  • Flow Cytometry analysis of Hela cells labelling RTN4/NOGO (red) with Ozanezumab (anti-RTN4/NOGO) (HY-P99213). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then cells were stained with the primary antibody at 1/200 dilution for an hour at 4℃. Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG Isotype Control (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
