Phytolacca americana Lectin (PWM) is an acidic monomeric glycoprotein specific to N-acetylglucosamine and a B cell-specific stimulator. Phytolacca americana Lectin acts as a mitogen to induce cell mitosis. Phytolacca americana Lectin sensitizes B cells to Toll-like receptor stimulation, enabling their activation by immunostimulatory DNA. When used in combination with copurified microbial Toll-like receptor ligands, Phytolacca americana Lectin triggers B cell proliferation and immunoglobulin secretion.

Description

Phytolacca americana Lectin (PWM) is an acidic monomeric glycoprotein specific to N-acetylglucosamine and a B cell-specific stimulator. Phytolacca americana Lectin acts as a mitogen to induce cell mitosis. Phytolacca americana Lectin sensitizes B cells to Toll-like receptor stimulation, enabling their activation by immunostimulatory DNA. When used in combination with copurified microbial Toll-like receptor ligands, Phytolacca americana Lectin triggers B cell proliferation and immunoglobulin secretion[1][2][3].

IC50 & Target[3]

IL-8

In Vitro

Phytolacca americana Lectin induces mitosis in peripheral lymphocytes through cell surface binding[2].
Phytolacca americana Lectin (1-10 μg/mL) induces the proliferation of live-gated CD19+ peripheral blood B cells and promotes plasmablast differentiation, while B cell receptor cross-linking via anti-Ig interferes with these effector functions[3].
Phytolacca americana Lectin significantly induces the expression of intracellular TNF in monocytes, as detected by mean fluorescence intensity[3].
Phytolacca americana Lectin (10 μg/mL; 24 h) induces IL-8 secretion in HEK293 cells transfected with TLR2, and this activity is partially dependent on TLR2[3].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

ELISA Assay[3]

Cell Line: human TLR2-transfected HEK293 cells
Concentration: 10 μg/mL
Incubation Time: 24 h
Result: Induced IL-8 secretion in TLR2-transfected HEK293 cells, with production almost doubled compared to non-transfected cells.
Reduced PWM-induced IL-8 production by approximately 50% (p=0.03) compared to isotype control after pre-treatment with 10 μg/mL anti-TLR2 mAb.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
