Description

Py-FGGK is a gelatinase-responsive short peptide. Py-FGGK can specifically target MRSA, cause leakage of cellular components, generate ROS in situ, and kill biofilm-encapsulated MRSA cells. Py-FGGK can promote cell migration to aid wound healing, and accelerates wound healing in MRSA-infected rat models. Py-FGGK can be used in studies related to MRSA infection[1].

In Vitro

The critical aggregation concentration of Py-FGGK in PBS aqueous buffer is 63.1 μM, and this concentration decreases to 39.8 μM after treatment with gelatinase, thereby promoting fibril formation[1].
Py-FGGK (300 μM; 0-24 h) adopts a random coil conformation in aqueous PBS solution, and transforms into an α-helical structure after incubation with 0.5 μg/mL gelatinase for 6 to 24 h, which corresponds to its cleavage into Py-FG[1].
Py-FGGK binds to HPS with moderate affinity, with a Kd value of 32.47 μM, and this binding process is mediated by its KKQK motif[1].
Py-FGGK potently inhibits the growth of MRSA with an MIC of 1 μM, and it kills MRSA with an MBC of 10 μM[1].
Py-FGGK (0.5-2 μM; 15 min-4 h) rapidly depolarizes the cell membrane of MRSA, induces significant leakage of cellular components from MRSA, and stimulates reactive oxygen species production in MRSA[1].
