Description

SBFI-AM is a Na+ selective fluorescent indicator. SBFI-AM shows selectivity for Na+ over K+ with Kd values of 6.7 and 166 mM, respectively (Ex = 340/380 nm; Em = 500 nm)[1].

In Vitro

Guide (The following are our recommended protocols; these are Guide only and should be modified according to your specific needs).
1. Preparation of Working Solution
This product has poor water solubility. Before adding it to cell culture medium or PBS buffer, mix an equal volume of 20-25% (w/v) Pluronic F-127 solution with the indicator stock solution in DMSO. Prepare a 1-10 μM working solution.
Note: Adjust the working solution concentration according to your actual situation and prepare fresh before use.

2. Cell Staining
2.1 Add an appropriate amount of dye working solution to the cells and incubate at room temperature for 40 minutes to 4 hours.
2.2 Observe using a fluorescence microscope or confocal laser scanning microscope.
Note: Since the Kd value of the indicator may differ between intracellular and solution, the intracellular SBFI should be calibrated using the pore-forming antibiotic Gramicidin (HY-P0163).
