YS-01 is an inhibitor of pendrin (SLC26A4) with pulmonary protective activity. YS-01 inhibits pendrin-mediated anion exchange activity, reduces pendrin expression, and suppresses the activation of NF-κB and the production of proinflammatory cytokines. YS-01 alleviates lipopolysaccharide-induced and ventilator-induced lung injury. YS-01 attenuates ovalbumin-induced airway hyperresponsiveness, inflammatory infiltration, epithelial thickening and goblet cell hyperplasia, and reduces eosinophil and neutrophil counts. YS-01 decreases MUC5AC transcription levels induced by IL-4, reverses the reduction of airway surface liquid volume, and inhibits the increase of apical SCN − concentration in epithelial cells. YS-01 can be used in studies related to lung injury and allergic asthma .

(S)-Lercanidipine is a calcium channel blocker with antihypertensive and neuroprotective activities. (S)-Lercanidipine reduces oxidative stress and protects auditory sensory hair cells from noise-induced damage. (S)-Lercanidipine showed significant protective effects on cell viability in in vitro experiments, especially at low concentrations. (S)-Lercanidipine has been found to effectively reduce the hearing threshold of mice after noise stimulation and protect the survival of outer hair cells. The antioxidant properties of (S)-Lercanidipine are reflected in its increased expression of antioxidant enzyme genes and decreased expression of oxidative enzyme genes. (S)-Lercanidipine administration is effective in reducing noise-induced hearing loss both before and after noise exposure. (S)-Lercanidipine may alleviate noise-induced hearing loss and protect the survival of outer hair cells through its antioxidant mechanism .

(1R)-cis-Bifenthrin is a widely used pyrethroid pesticide with activity that reduces motor coordination. (1R)-cis-Bifenthrin has a significant impact on the motor function of ParKin-/- mice, as shown by increased pole climbing time and wheel running Time decreases. Exposure of (1R)-cis-Bifenthrin resulted in a significant reduction in tyrosine hydroxylase-positive cell counts and protein expression. (1R)-cis-Bifenthrin caused increased expression of mitophagy-related proteins LC3B and p62. (1R)-cis-Bifenthrin has a lower binding energy with transferrin and transferrin receptor 2, showing stronger interactions. The biological effects of (1R)-cis-Bifenthrin show relationships with mitophagy and ferroptosis-related pathways .

Inflammation/Immunology

LW-1 is a fluorescent probe targeting polarity detection of lipid droplets (LD). LW-1 produces a strong solvatochromic effect via the intramolecular charge transfer (ICT) mechanism: as environmental polarity increases, the emission wavelength red-shifts and the fluorescence intensity decreases significantly (emission at 557 nm in toluene and 642 nm in acetonitrile, with an 85 nm red-shift); fluorescence enhances in low-polarity lipid droplets. LW-1 can be used for lipid droplet polarity detection, differentiation between cancer cells and normal cells, monitoring of lipid droplet polarity changes under different physiological conditions such as starvation, oleic acid treatment and oxidative stress, as well as ex vivo imaging of polarity changes in lung tissues of asthmatic mice. The detection wavelength for cell imaging with LW-1 is Ex/Em = 405/500-550 nm (green channel) .

Nile-TPA is a fluorescent probe for polarity detection, which can be used for lipid droplet targeting, cell polarity analysis, in vivo polarity imaging, and blood polarity monitoring in circulating tumor mouse models. The detection mechanism of Nile-TPA relies on the dual intramolecular charge transfer (ICT) effect generated by the electron-donating triphenylamine and Nile Red moieties and the electron-withdrawing carbonyl group; this probe emits strong red fluorescence in a low-polarity environment, and as polarity increases, the fluorescence intensity decreases and a red shift occurs due to changes in the excited-state dipole moment. In spectral tests, its excitation wavelength is 540 nm, and the emission wavelength varies in the range of 550-800 nm depending on solvent polarity; for cell, zebrafish and mouse imaging, Ex/Em = 561/570-620 nm, and for in vivo mouse imaging, Ex/Em = 560/620 nm .

P2-Aggregate is a polarity-sensitive fluorescent probe for protein aggregation, which is based on the Nile Red scaffold and modified with a morpholine ring to enhance water solubility. P2-Aggregate improves anti-precipitation ability in aqueous phase via morpholine ring modification, and enables site-specific labeling of target proteins by combining with HaloTag technology. Utilizing the property that its emission wavelength blue-shifts with decreasing local polarity (excited at 543 nm), P2-Aggregate quantitatively detects changes in local polarity during protein aggregation and polarity heterogeneity inside aggregates in living cells. P2-Aggregate can be used in the research of diseases associated with the aggregation of Htt-110Q and SOD1 proteins, such as Huntington's disease and amyotrophic lateral sclerosis .
