Simridarlimab

Simridarlimab (IBI-322) is a bispecific antibody targeting PD-L1 and CD47. Simridarlimab attenuates CD47 activity in monovalent binding and blockes PD-L1 activity in bivalent binding. Simridarlimab selectively binds to CD47+PD-L1+ tumor cells, effectively inhibits CD47-SIRPα signal and triggered strong tumor cell phagocytosis in vitro.

Immobilized CD47 Protein, Human (HEK293, HY-P72917) can bind Simridarlimab. The EC50 for this effect is 134.7 ng/mL. -
Immobilized PD-L1 Protein, Human (HEK293, HY-P73361) can bind Simridarlimab. The EC50 for this effect is 188.8 ng/mL. -
Flow cytometric analysis of 1×106 Jurkat cells labelling CD47 (red) with Simridarlimab (HY-P99903). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa Isotype Control (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
