STAT6-IN-14 (Compound 39) inhibits the binding of human STAT6 to phosphorylated IL-4 receptor α (pIL-4Rα) in a cell‑free system, with an IC₅₀of 1.20 μM (30 min pre‑incubation + 30 min fluorescence polarization detection)^[1].
STAT6-IN-14 (0.1-1 μM; 1 h pre‑incubation + 6 h IL‑4 stimulation) inhibits STAT6‑mediated transcriptional activity in HEK293 cells overexpressing human STAT6, with inhibition rates of 99% at 1 μM and 84% at 0.1 μM, and no significant cytotoxicity is observed^[1].
