In Vitro

T7117 (1-10 mM; 5 days) inhibits the proliferation of U251 and U118 glioblastoma cells[1].
T7117 disrupts the TRIM25-HIF-1α interaction in U251, U118, and 293T cells, inhibits TRIM25-mediated HIF-1α stabilization and ubiquitination in a K532-dependent manner, and reduces HIF-1α protein levels in U251 and U118 glioblastoma cells[1].

Result: Significantly inhibited the proliferation of U251 and U118 GBM cells at 1 mM.
Exerted significant inhibitory effects on GBM cell proliferation at 10 mM, with effects comparable to those of T17056 and greater than other tested compounds.
In Vivo

T7117 (10 μg; i.p.; every 2 days for 35 days) inhibits intracranial glioblastoma growth and prolongs survival in tumor-bearing BALB/c nude mice without significant toxicity[1].

Animal Model: BALB/c nude (4-week-old female, 12-16g, intracranial implantation of 5×105 glioblastoma cells)[1]
Dosage: 10 μg
Administration: i.p.; every 2 days
Result: Inhibited intracranial tumor growth as measured by bioluminescent imaging.
Prolonged survival time of mice bearing luciferase-labeled U251 tumors.
Reduced tumor cell proliferation as shown by Ki-67 staining of tumor sections.
Caused no significant treatment-related toxicity, with stable body weight and no histopathological abnormalities in liver, lung, and kidney.
