Description

Trecovirsen is an antiviral agent targeting HIV gag mRNA, which hybridizes with complementary HIV gag mRNA at the initiation site. Trecovirsen induces a reversible, dose-dependent prolongation of activated partial thromboplastin time via its polyanionic properties. Trecovirsen is applicable to research related to HIV infection[1][2][3].

In Vitro

Trecovirsen produces potent, long-lasting inhibitory effects against HIV in cultured HIV-infected cells[1].
Trecovirsen (GEM91) (0.2 μM) potently inhibits HIV-1 replication in multiple in vitro cell models, including acutely and chronically infected CEM cells, human peripheral blood lymphocytes, macrophages, and cells infected with clinical HIV-1 isolates, with an IC50 of 0.2 μM[3].

In Vivo

Trecovirsen (4 mg/kg; i.v. infusion; over 2 hours) administered via 2-hour intravenous infusion in cynomolgus monkeys results in widespread tissue distribution (with highest accumulation in liver and kidney), a mean plasma elimination half-life of 55.8 hours, a mean blood elimination half-life of 75.4 hours, and excretion of 70% of the dose in urine over 240 hours, with both intact drug and shorter-length metabolites detected in plasma and tissues[2].
Trecovirsen (GEM91) (30 mg/kg; i.v.; single bolus injection) exhibits a short distribution half-life (0.95 hr) and prolonged elimination half-life (47.57 hr) after single i.v. bolus administration at 30 mg/kg in male Sprague-Dawley rats, with wide tissue distribution, preferential retention in kidney, liver, and thyroid, and urinary excretion as the primary elimination pathway[3].
