XAT-14 is a Furanocoumarin derivative. XAT-14 competes with agonists to bind to the active pocket of MRGPRX2, inhibiting calcium influx and mast cell degranulation. XAT-14 suppresses the release of histamine, IL-8 and MCP-1 induced by Compound 48/80 (HY-115768). XAT-14 inhibits local allergic inflammation. XAT-14 can be used for the research of pseudo-allergic diseases.

XAT-14 (100 μM) shows no cytotoxicity in LAD2 cells[1].
XAT-14 (25-100 μM) potently inhibits Compound 48/80-induced β-hexosaminidase release in LAD2 cells, with an IC50 of 22.95 μM, and exhibits superior efficacy to Imperatorin (HY-N0285)[1].
XAT-14 (25-100 μM) dose-dependently inhibits Compound 48/80-induced release of histamine, IL-8 and MCP-1 in LAD2 cells[1].
XAT-14 (25-100 μM) inhibits Compound 48/80-induced calcium influx in MRGPRX2-HEK293 cells in a dose-dependent manner, indicating that its effect is mediated through the MRGPRX2 signaling pathway[1].

- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

This protocol yields a suspended solution of 5 mg/mL. Suspended solution can be used for oral and intraperitoneal injection.
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
