Description

Y502-3888 is a c-Myc inhibitor. Y502-3888 binds to the c-Myc G4 structure, inhibits c-Myc transcription, and downregulates c-Myc expression at both mRNA and protein levels. Y502-3888 inhibits the viability of myeloma cells and induces cell apoptosis. Y502-3888 can be used for the research of multiple myeloma[1].

In Vitro

Y502-3888 (1-30 μM; 48 h) potently and selectively inhibits the viability of human multiple myeloma RPMI 8226 and U266 cells, with IC50 values of 12.67 μM and 7.03 μM, respectively, and minimal toxicity to non-malignant HEK293T cells[1].
Y502-3888 (5-20 μM; 48 h) dose-dependently induces apoptosis in human multiple myeloma U266 cells, with increasing proportions of early and late apoptotic cells at concentrations of 5, 10, and 20 μM[1].
Y502-3888 (5-20 μM; 48 h) dose-dependently downregulates c-Myc protein expression in human multiple myeloma U266 cells[1].

Apoptosis Analysis[1]

Cell Line: Human multiple myeloma U266 cells
Concentration: 5, 10 and 20 μM
Incubation Time: 48 h
Result: Induced dose-dependent increases in apoptotic cell populations.Resulted in 8.68% early apoptotic and 15.4% late apoptotic cells at 5 μM.
Resulted in 16.2% early apoptotic and 26.2% late apoptotic cells at 10 μM.
Resulted in 24.9% early apoptotic and 57.0% late apoptotic cells at 20 μM.

Western Blot Analysis[1]

Cell Line: Human multiple myeloma U266 cells
Concentration: 5, 10 and 20 μM
Incubation Time: 48 h
Result: Reduced c-Myc protein levels in a dose-dependent manner.
Caused statistically significant reductions in c-Myc protein levels at 10 μM and 20 μM relative to control cells.
Markedly decreased c-Myc protein levels at 20 μM.

Real Time qPCR[1]

Cell Line: Human multiple myeloma U266 cells, CA46 Burkitt's lymphoma cells
Concentration: 5, 10 and 20 μM
Incubation Time: 48 h
Result: Reduced c-Myc mRNA levels in U266 cells in a dose-dependent manner, with expression reduced to ~50% of control at 20 μM.
Dose-dependently suppressed Exon 1 c-Myc mRNA levels in CA46 cells, with minimal effect on Exon 2 levels.
