5-FITC tyramide is a green fluorescent dye composed of FITC (HY-66019) and tyramide (Ex=488 nm, Em=525 nm). 5-FITC tyramide is utilized as reporter fluorescent substrate of horseradish peroxidase (HRP)-catalyzed deposition for tyramide signal amplification (TSA). 5-FITC tyramide can be used for multiplex immunohistochemistry (mIHC).

Biological Activity
Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
5-FITC tyramide assay
1. Inject into the rat ganglion with 4 μL of WGA-HRP tracer, and then get specific tissues for slide.
2. Slides were allowed to dry for 60 min.
3. Rehydrate in TNT buffer (0.1 M Tris-HCl, 0.15 M NaCl, 0.05% Tween 20, pH 7.5), and then quench in freshly prepared NaBH4 (0.5 mg/mL) in TN buffer (0.1 M Tris-HCl, 0.15 M NaCl, pH 7.5) for 15 min.
4. Wash in TNT and then permeabilized in 0.1% Triton X-100 in TN for 15 min.
5. Incubate in TNT plus 2% BSA (TNT-BSA) for 60 min following washing in TNT, and then incubate min in 5-FITC tyramide for 15 min (1: 50 with amplification diluent).
6. Wash in TNT, and then apply the primary antibodies overnight at 4°C following detecting by incubation with donkey anti-rabbit IgG conjugated to Lissamine rhodamine (LRSC) or Cy3 (HY-D0822) (1:200 with TNT-BSA) and donkey anti-sheep Cy5 (HY-D0821) (1:200 with TNT-BSA).
7. Fluorescence microscopy detection.
Note: it is suggested that the optimal concentration of 5-FITC tyramide working solution according to actual conditions.
