Description

6-ROX is a selective fluorescent probe and potential inhibitor of COX-2. 6-ROX binds to the active site of COX-2 and inhibits its conversion of arachidonic acid into prostaglandins. 6-ROX is often used in the field of optical imaging related to tumors and inflammation, and helps detect diseased tissues with high expression of COX-2[1][2].

In Vitro

6-ROX can be conjugated to COX1/2 inhibitors such as Indomethacin (HY-14397). The 6-ROX conjugate can effectively cross the cell membrane to inhibit COX-2, and exhibits COX-2 inhibitory activity in LPS-activated RAW264.7 cells and 1483 HNSCC cells[1].
In in vitro cell optical imaging experiments (200 nM; 30 min treatment, 30 min washout),the 6-ROX conjugate can induce strong red fluorescence in 1483 HNSCC cells that highly express COX-2, and this uptake and labeling can be blocked by the COX inhibitor Indomethacin (5 μM, 10 μM; 20 min) [1].
