8-Oxo-GTP is an oxidized guanine nucleotide, produced by reactive oxygen species (ROS). 8-Oxo-GTP serves as a substrate for MutT protein, GTP cyclohydrolase II, Mycobacterium tuberculosis MutT1, and Mycobacterium tuberculosis Rv1700, which hydrolyze it to corresponding nucleoside monophosphates or diphosphates. 8-Oxo-GTP causes detrimental effects including mutations and mistranslation when incorporated into nucleic acids. 8-Oxo-GTP can be used for the research of cancer and tuberculosis[1][2][3].

In Vitro

8-Oxo-GTP (8-Hydroxyguanosine triphosphate) (15-100 μM; 30 min) is efficiently hydrolyzed to 8-Oxo-GMP by purified and histidine-tagged recombinant Escherichia coli GTP cyclohydrolase II with an apparent Km of 21 μM and a Kcat of 4.0 min-1, at a rate higher than that of GTP hydrolysis[1].
8-Oxo-GTP (250 μM; 30 min) is converted to 8-oxo-GDP by purified MtuMutT1 at 37 °C[2].
8-Oxo-GTP (250 μM; 30 min) is converted to 8-oxo-GDP and 8-oxo-GMP by purified MtuMutT1 and Rv1700 together at 37 °C[2].
