Biological Activity
AChRα(97-116) (10 μg/mL; 72 h) does not induce significant proliferation of lymphocytes harvested from MBP68-86-immunized Lewis rats[1].
AChRα(97-116) (10 μg/mL; 48 h) maintains the antigen-specific immune cell phenotype responsive to dopaminergic modulation in recall-stimulated EAMG rat-derived CD4+ T cells and B220+ B cells[2].
AChRα(97-116) (2 μg/mL; overnight at 4 °C) effectively detects elevated AChR-specific antibody levels in EAMG rat serum and dopamine-stimulated EAMG rat splenocyte supernatants[2].
AChRα(97-116) (10 μg/mL; 24 h pre-incubation + 24 h co-culture) stimulates CD4+ T cells from EAMG rats to differentiate into AChR-specific Tfh cells at a 3-4-fold higher ratio than in control rats, and these Tfh cells promote increased IgG2b-secreting B cell percentages and anti-AChR antibody production in co-cultures with EAMG-derived B cells[3].
AChRα(97-116) (10 μg/mL; 72 h) stimulates lymph node-derived mononuclear cells from EAMG rats to produce elevated percentages of CD4+CXCR5+ and CD4+ICOS+ pre-Tfh cells following 72 h of incubation[3].
AChRα(97-116) (2 μg/mL; overnight at 4°C) coated on ELISA plates enables detection of significantly elevated anti-AChR antibody levels in serum from EAMG rats across multiple time points and dilutions compared to control rats[3].

Immunization with AChRα(97-116) (50 μg; subcutaneous injection; day 0 and day 30) induces experimental autoimmune myasthenia gravis in Lewis rats with >85% efficiency, driving elevated anti-AChR antibody production, reduced muscle AChR clusters, and expansion of AChR-specific pre-Tfh and mature Tfh cells that promote germinal center formation and B cell antibody secretion[3].

This protocol yields a suspended solution of 2.22 mg/mL. Suspended solution can be used for oral and intraperitoneal injection.
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (22.2 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
