Description

Antibacterial agent 351 is a broad-spectrum antibacterial agent. Antibacterial agent 351 binds to the allosteric site of PBP2a, induces conformational changes, and reduces the level of PBP2a in MRSA. Antibacterial agent 351 acts as a DNA intercalator to interfere with bacterial DNA function. In Vitro

Antibacterial agent 351 (0.25 × MIC - 8 × MIC) inhibits MRSA virulence factors in a dose-dependent manner, reducing auto-aggregation, decreasing hydrophobicity by ~18.6% at 0.5 × MIC, lowering EPS content to 62.5% of the control at 2 × MIC, and eradicating up to 35% of mature MRSA biofilm at 8 × MIC[1].

In Vivo

Antibacterial agent 351 (10 × MIC) exhibits potent in vivo anti-MRSA activity. When administered at 10 × MIC, it increases the 7-day survival rate of Galleria mellonella (greater wax moth) larvae infected with MRSA to 70%[1].
Antibacterial agent 351 (50 μg/mL) shows no acute toxicity to Galleria mellonella larvae when administered at a dose of 50 μg/mL, with a 100% survival rate within 5 days[1].
