DEPMPO

DEPMPO is a phosphorus-substituted nitrone spin trap probe that can capture superoxide anions and hydroxyl radicals. DEPMPO forms stable spin adducts with hydroxyl, aryl, sulfite and superoxide radicals, and the formation of these adducts can be inhibited by catalase or superoxide dismutase. DEPMPO is rapidly absorbed after intraperitoneal injection and distributes uniformly in the liver, heart and blood of mice. DEPMPO can be used in studies related to oxygen radical detection.

In Vitro
DEPMPO displays concentration- and solvent-dependent radical trapping profiles; at 25 mM, only carbon-centered radicals generated by the reduction of compounds 1 and 2 are trapped; at 100/105 mM, ・OH can be captured additionally; at 250 mM, aryl, carbon-centered, and ・OH radicals from compound 2 reduction are simultaneously detected. When methanol or DMSO is supplemented into the reaction system, DEPMPO only traps solvent-originated or carbon-centered radicals[2].
DEPMPO (1 μM-100 mM, 16 h incubation) displays dose-dependent cytotoxicity toward CHO; concentrations ≤25 mM cause negligible cellular damage, 50 mM drastically reduces colony formation, and 100 mM completely abolishes colony growth[3].
DEPMPO (30 mM; decay monitored over time) forms the SO3•− spin adduct, which has a lifetime of 136 minutes in PBS in vitro and 22 minutes in the presence of 2 mM ascorbic acid[3].
DEPMPO solutions prepared in pure H2O or phosphate buffer at room temperature remain stable without artifactual nitroxide generation; DEPMPO efficiently captures ·OH and superoxide radicals, with respective second-order rate constants of 7.8×109 dm3 mol-1 s-1 and 90 dm3 mol-1 s-1[5].
DEPMPO-OH remains stable under GSH exposure yet undergoes rapid decomposition by ascorbic acid, whereas DEPMPO-OOH is degraded by both ascorbic acid and GSH.[4].
DEPMPO exhibits lower efficiency than CYPMPO in capturing superoxide anions in human blood polymorphonuclear leukocytes stimulated by PMA (HY-18739)[4].

- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 5 mg/mL (21.26 mM); Clear solution
This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 5 mg/mL (21.26 mM); Clear solution
This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
