In Vitro
ATR-IN-1 (Compound 13) inhibits ATR with an IC50 of 0.0005 µM and shows selectivity over ATM (IC50 15 µM) and DNAPK (IC50 >25 µM)[1].
ATR-IN-1 (1 h) does not inhibit mTOR signaling in TSC1−/− MEF cells (IC50 >13 µM)[1].
ATR-IN-1 has a hERG IC50 >30 µM by automated whole-cell electrophysiology[1].
ATR-IN-1 does not inhibit CYP3A4, 2D6, 2C9, or 1A2 (IC50 >24 µM)[1].
ATR-IN-1 (72 h) inhibits the growth of ATM-null HT144 melanoma cells with a GI50 of 0.35 µM after 72 h continuous exposure[1].

Cell Cytotoxicity Assay
Cytotoxicity assays are usually based on the assessment of cell membrane damage, which can also be indirectly detected by measuring cell viability. Detection methods include MTT assay, CKK-8 assay, LDH assay and ATP assay, etc.
