Description

BXY-14 is a TLR2 agonist and vaccine adjuvant. BXY-14 significantly downregulates the expression of intratumoral PD-L1 in mouse models. BXY-14 acts as a vaccine adjuvant to induce antibody responses. In Vitro

BXY-14 (1-106 pM) acts as a potent TLR2 agonist in hTHP-1 cells, inducing TNF-α release with an EC50 of 2.2 nM[1].
BXY-14 (10-3-103 nM; 24 h) is a potent TLR2 agonist in hPBMC that induces TNF-α release with an EC50 of 0.58 nM[1].

In Vivo

BXY-14 (10-20 mg/kg; s.c.; on days 0, 14, and 28) functions as a potent vaccine adjuvant in mice, inducing stronger OVA-specific antibody responses with a Th1-skewed bias compared to Diprovocim, and is well-tolerated with no adverse body weight effects[1].
BXY-14 (10 mg/kg; s.c.; on days 0, 14, and 28) in combination with anti-PD-L1 enhances tumor growth inhibition, prolongs survival to 100% at day 50, and reduces intratumoral PD-L1 expression in a murine B16-OVA melanoma model, while BXY-14 (20 mg/kg; s.c.; on days 0, 14, and 28) as monotherapy also suppresses tumor growth and downregulates PD-L1[1].
